B. aristata i.e. Daruharidra is officially noted in, Indian pharmacopeia, ayurvedic and siddha pharmacopoeia of India. Due to its various proven pharmacological activities it is an important part of polyherbal formulations in the treatment of different diseases and disorders. Berberis aristata is the main constituents of rasaut, darvyadi kvatha, darvyadi leha, darvyadi taila, rasanjana, dasanga lepa. Aromoline, oxyberberine, oxyacanthine, berbamine, berberine, pakistanine, 1-O-methylpakistanine, pseudopalmatine chloride are the main chemical constituents. Anti-diarrheal activity, Cardiotonic activity, Hepatoprotective activity anti-diabetic activity and anti-cancer activity are reported. With reverse pharmacology and their scientifically validation, as a result more than 30 patents are filled on different formulation of B.aristata and granted. With this background, bark of B. aristata was selected for the validation and standardisation of previous studies. Berberine chloride as standard 1 mcg/µl and 1 mg/ml sample of Berberis aristata were prepared. CAMAG Automatic TLC Sampler 4 (ATS4) "ATS4_201152" S/N 201152 (1.02.18) is used for the identification using the D2 Lamp at wavelength 254 nm. Preliminary phytochemical analysis of Berberis aristata showed the presence of carbohydrate, glycoside, alkaloid, protein, amino acid, saponin, tannin and flavonoid. Microbial limits were found within permissible limits. HPTLC analysis of the methanolic extract of crude drug showed the presence of berberine in qualitative and quantitative 90 ng in 1 µl at a λmax 346.
